    【Abstract】 Objective To investigate the apoptosis of human erythroleukemia K562cells and identify the difˉference in gene expression profile of K562cells induced by sodium cantharidate.Methods Saline as negative conˉtrol,sodium cantharidate of different concentration was added to K562cells,then MTT assay and DNA gel elecˉtrophoresis were carried out to detect thecellapoptosis.ThePCRamplifiedcDNAwerespotteonpoly-L-lysine-coatedmicroscopeslidesbycapillaryactionusingstainlesssteelpinstoproducea30×18microarrays.Total RNA was extracted by using Trizol reagent,40μg of total RNA labeled by reverse transcription using SuperscriptⅡtranscripˉtase,oligo(dT)primer,and Cy5-dCTP orCy3-dCTP for experimental and control samples,respectively.Then the labeled cDNAs were hybridzed to microarrays at42℃overnight and were imaged by using a dual-laser scanner.The experimental data were calculated with the Quantarray software package.Results Significant growth inhibition ocˉcurred in K562cells in vitro after sodium cantharidate treated ......
